Abstract
Summary. Introduction. Chronic hyperglycemia in patients with diabetes mellitus (DM) leads to cardiovascular complications, which are
based on disorders of the vascular-platelet component of hemostasis. Therefore, it is advisable to evaluate the effect of antihyperglycemic
agents on platelet function and to develop alternative methods for its correction. Aim: to examine the effect of the combined use of skin
flap autologous grafting (SFAG) and liraglutide on the vascular-platelet mechanism of hemostasis in white rats with absolute insulin deficiency caused by alloxan-induced DM. Materials and Methods. The study was performed on white rats divided into five groups: control (intact
animals), comparison (rats with alloxan-induced DM), and experimental groups1, 2, and 3 (rats with alloxan-induced DM who were administered liraglutide at 0.4 mg/kg/day, underwent SFAG, and received a combination of SFAG and liraglutide at 0.4 mg/kg/day, respectively). Intraperitoneal administration of a 5% alloxan solution was used to model DM. To assess the state of the vascular-platelet mechanism of
hemostasis, platelet aggregation activity was evaluated, and the concentration of blood endothelial dysfunction (ED) markers, such as
syndecan‑1, soluble E-selectin, (sE-selectin), and asymmetric dimethylarginine (ADMA) was determined. Results. Course administration of
liraglutide to rats with DM normalizes carbohydrate metabolism, partially restores platelet aggregation activity, and reduces the severity
of inflammatory activation of the endothelium. Isolated SFAG also normalizes carbohydrate metabolism, partially restores the functional
activity of platelets, but does not cause a complete recovery of the endothelium structural and functional state. The combination treatment approach helps normalize carbohydrate metabolism, completely restores platelet aggregation activity, and reduces the severity of ED.
Conclusion. The combination of SFAG and a course of liraglutide in rats with alloxan-induced DM is superior to each of the methods individually in terms of the effective correction of platelet aggregation activity, as well as recovery of the endothelium structure and function.
